Adriamycine. Health-based calculated occupational cancer risk values

Authors' recommendations: At the request of the Minister of Social Affairs and Employment, the Dutch Expert Committee on Occupational Safety (DECOS), a committee of the Health Council of the Netherlands, derives so-called health-based calculated – occupational cancer risk values (HBC-OCRVs) associated with excess cancer levels of 4 per 1,000 and 4 per 100,000 as a result of working life exposure to substances. It concerns substances which are classified by the Health Council or the European Union in category 1A or 1B, and which are considered stochastic genotoxic carcinogens. For the estimation, the Committee uses the Guideline for calculating carcinogenic risks of the Health Council.1 In this report the Committee evaluates the possibility to establish such estimates for adriamycin. Adriamycin is a cytotoxic anthracycline antibiotic used in antimitotic chemotherapy. In this report, the Committee concludes that adriamycin is a carcinogenic substance with a stochastic genotoxic mechanism. The Committee recommends adriamycin to be classified in category 1B (substance presumed to be carcinogenic to humans). The Committee is of the opinion that due to a lack of sufficient data, it is not possible to estimate the additional lifetime cancer risk for adriamycin.
