The chemotherapy drug, 5-fluorouracil (5-FU), is used for a wide variety of tumor types. However, there are many factors affecting its conversion to an active form and elimination from the body, which makes it difficult to select the optimal dose of this agent for individual patients. Pharmacokinetically (PK) guided dose adjustment involves repeated measurement of 5-FU and/or its metabolites in the bloodstream of the patient and modification of the 5-FU dose based on these measurements to select the dose that provides the most favorable balance of antitumor efficacy
versus drug toxicity for each patient.
